Daily Challenge is a special mode in Bloons Tower Defense 5 that was added in an update on Jan 27, 2012. The player must beat the pre-defined track with some special conditions, like a restriction to only certain types of towers, road items, upgrades or custom RBE. The Daily Challenge changes at midnight (GMT/UTC). There is a similar mode in Bloons TD 5 Deluxe and Bloons TD 5 iOS, and it is called Random Challenge.

The player is rewarded with Monkey Money for completing the Daily Challenge and also gets a Medal and Awesome Points for the specific number of challenges completed. A standard reward of $50 Monkey Money is usually given upon completion. Player is rewarded with full Monkey Money reward for Today's Daily Challenge once a day and with 40% of original reward, if the challenge is repeated. Also, the player can finish Daily Challenge Vault challenges for 40% reward and repeat them for 16% (40% of 40%). This does not apply to all of the Daily Challenge Vault challenges, as the "ZOMG", "20 Ceramic bloons on the wall", "Don't Mess Up!", and "That Was Quick" challenges have been reduced to $2.

- The player must be registered and logged in with a Ninja Kiwi account to attempt Daily Challenges.
- Sometimes in general, the greater the Monkey Money reward, the harder the challenge is.
- Also, the Monkey Money reward may depend on the length of the challenge.
- The game cannot be saved during Daily Challenges.
- At least Rank 19 is required to play Daily Challenges.
- The Nature Calls daily challenge is the first Daily Challenge where the player gets special agents for free.
- Completing a map for a Daily Challenge, will not earn the medal for the map.
- The Daily Challenge Vault allows previous Daily Challenges, mostly for a 40% reward.
- The daily challenge from 26th Mar, 12 means B-Day. This word is played on the real D-Day.
- The achievement, Otherwordly (gained by completing 365 sequential first run Daily Challenges) was removed beacause it's nearly impossible for any player to ever earn it.
- On the January 10, 2013 challenge, there are actually a lot of other upgrades to pop Lead Bloons, since all attacking towers have at least one upgrade which allows them to pop lead.
- On March 13, 2012, it says to complete the track Park Path, but in Monkey Lane is the actual track.
- On May 28, 2012, custom Bloons waves were introduced in Daily Challenges.
- On the DC "Precision Pineapples", both x/3 Super Monkey and x/3 Sniper can pop leads, and are allowed.
- Daily Challenges 50 and 71 both have the same name, "Icebreaker." However, the actual challenges are different.
- Daily Challenge 12, which is called "Beat 10 Epic Rounds with Lots o' Cash" actually features 11 rounds.
- Limits to the number of Towers and Special Agents at a time or the number of Towers and Special Agents type don't apply to Super Monkey Storms.
- The 20 Ceramic Bloons on the Wall challenge can be beaten by spending all the provided cash on road spikes.
- The August 26th Daily Challenge gives 1/2 as much Monkey Money on the first time winning as the first time winning Castle on Hard, if Castle Hard isn't completed with double Monkey Money.
- On the Strapped for Cash challenge (Febuary 11, 2012's challenge), it is actually possible to get through the early rounds without using Special Agents by using the Dart Training Facility, to earn a free dart monkey.
- In the September 20 challenge (Camo, Right?), the only thing that can pop Camo Bloons is the Dartling Gun, unless Tier 3 Pirate Cove or Tier 3 Super Monkey Lair is active.
- In Daily Challenge Precision Pineapples nothing that can pop lead bloons can be used except Pineapples, making it a pretty hard challenge.
- Some challenges are played on the films or things in real life, such as in film 'Kill Bill' (it was from July 5 2012), D-Day (26th Mar 2012) or PSY's Gangnam Style (13 December).
- In the DC "Castle of Nobility", it accidentally started on Monkey Lane. Same with December 16 2012's challenge.
- The DC "Floating Downstream" was originally supposed to be played on Downstream, but was accidentally put on the field Snake River. Two days later they made the challenge "Rushing Downstream", which is basically the same challenge, except placed on the intended map: Downstream.
- The DC "Rushing Downstream" is the first DC on the Downstream map.
- The DC "Fives" was suggested by a user called "LemonadeFire". NinjaKiwi accidently posted "LemonadeFive" as the creator of the challenge.
- The DC "Set-up Chaos" originally had a upgrade restriction bug where the Monkey Buccaneer, Apprentice, and Village were restricted to 1/3 opposed to 3/1.
- The DC "I Spy With My Eye" is supposed to end on Round 70. When you first start the challenge, it says 85. It changes to 70 later on though.
- In the DC "Popping Evolved" (November 1 2012 challenge), the only unit that can pop Camo Lead Bloons is the Sniper Monkey, unless using Special Agents, Premium Upgrades or Tier 3 Super Monkey Lair on a X/3 Robo-Monkey.
- The Feb 8 2012 challenge is called "Bullet, Cannon and the BB" (Bionic Boomer), so the Glaive Lord should not be allowed in the challenge. However, without the use of premiums, Glaive Lord is the only way to pass round 47, because a 2/3 Sniper Monkey is not enough to pop all camo bloons in the level or survive with 100 lives. Because this, Glaive Lord can still be used in the challenge.

Here is a list of all Daily Challenges which have been available in BTD5. Only special conditions are mentioned with an exception towers and road items which are not mentioned are not available. So, if not specified:
- $50 Monkey Money as standard reward.
- Easy is from wave 1 to 50, 200 lives
- Medium is from wave 1 to 65, 150 lives
- Hard is from wave 1 to 85, 100 lives
- All Special Agents available for Monkey Money
- No Special Agents given for free
- Standard starting cash $650 ($50,000 in Deflation) (of course, double starting cash with Double Cash)
- Full upgrades available for towers unless specified (below)
- (Example 1) Dart Monkey (2-3) – means that max. 2nd upgrade in path 1 allowed and max 3rd upgrade in path 2 is available for the Dart Monkey.
- (Example 2) Dart Monkey (4-3) – means that max. 4th upgrade in path 1 allowed and max 3rd upgrade in path 2 is available for the Dart Monkey tower type, but not both for the same tower, so maximum for a tower is 4-2 or 2-3
